This cell lines features a stable knockout of PPOX-Cys459 in human U-2 OS cells , using CRISPR/Cas9 technology. Knocking out the Cys459 locus of the PPOX gene and effectively abolishing protoporphyrinogen oxidase activity creates a model that mirrors aspects of variegate porphyria, presenting with substantial accumulation of protoporphyrinogen IX and decreased heme levels, which in turn impacts the cell’s oxygen consumption rates and alters mitochondrial function. This cell line is pivotal for investigating the molecular and cellular consequences of disrupted heme synthesis, providing a direct link to studying diseases associated with heme biosynthesis defects, including porphyrias, and offering a platform for therapeutic exploration.
