PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) are recommended for optimal cell culture. DMEM/F-12 (1:1) Medium (TM510) + 10% FBS + 1% Penicillin/Streptomycin Solution (G255) ,37.0°C ,5% CO₂. 3.0 µg/ml Puromycin (G264) for selection. Note: Selection drugs should be added to the culture medium after the first passage to ensure cells have recovered from freeze-thaw conditions. |

MEF Psen1/2-/- cells were derived from E8.5 embryos which are neomycin and hygromycin-resistant and were immortalized with SV40 large T antigen via transfection with the pMSSVLT plasmid. Human PSEN1 D257A mutant expression was established through retroviral transduction with pMSCV puro hPSEN1WT plasmid with site directed mutagenesis and stable expression was determined via puromycin selection. PSEN1 (Presenilin 1) is a gene of interest in neuropathology and diseases such as Alzheimer’s disease (AD) in which patients with an inherited form of the disease carry mutations.
