Dulbecco's Modified Eagle Medium (DMEM) ,High Glucose (TM500) + 5% FBS + 2 mM L-Glutamine (G275) + 1% Penicillin/Streptomycin Solution (G255) ,37.0°C ,5% CO₂. 150 µg/ml Geneticin/G418 (G271) for selection. Note: Selection drugs should be added to the culture medium after the first passage to ensure cells have recovered from freeze-thaw conditions. |

MC38 murine colonic adenocarcinoma cells were transduced with a retrovirus containing the human MUC1 gene, a mucin family glycoprotein commonly associated with epithelial cancers. The resulting MUC1-positive cell line was selected by G418 resistance and FACS analysis. MUC1 is a cell surface glycoprotein known for its role in providing protective, lubricative barriers at epithelial surfaces and is also implicated in cellular signaling and tumor progression, making it a target of interest in oncology research.
