This cell line was generated by a CRISPR/Cas9 mediated knockout of NADH dehydrogenase [ubiquinone] iron-sulfur protein 2 (NDUFS2) – one of three protein subunits of interfacial region in human embryonic kidney cell line 293 (HEK293). Protein knockout was verified through Western blot analysis. This cell line, HEK293ΔNDUFS2, grows much slower than the parental cell line and requires adequate media change during growth. Additinally, HEK293ΔNDUFS2 cells show decreased Complex I specific respiration, glycolytic capacity, ATP pool and cell membrane integrity, but shows increased Complex II respiration, ROS generation, apoptosis and necrosis. Treatment with idebenone partially restores growth, ATP pool and oxygen consumption in the knockout cells. This cell line reveals the importance of NDUFS2 for cell growth and metabolism and is suitable for research evaluating the efficacy of mitochondrial therapeutics for bioenergetic capacity.
