T3489 (P301SV2H (NK23)) is one of two novel cell lines designed for the ultrasensitive detection of tau seeding activity, particularly in the context of Alzheimer’s disease research. These biosensor cell lines are engineered to express tau protein segments linked to fluorescent proteins, enabling fluorescence resonance energy transfer (FRET) based detection of tau aggregates. Each line offers unique features tailored to different research needs, enhancing both the sensitivity and specificity of tau pathology detection. Using a lentiviral vector, the tau protein segment (amino acids 246 to 378) with the P301S mutation was fused under a human cytomegalovirus (CMV) promoter to fluorescent proteins mCerulean3 or mClover3. The sequence linking the tau segment to the fluorescent proteins was optimized to improve FRET efficiency, essential for detecting tau aggregation dynamics.T3489 cells are engineered for high expression levels of tau fusion proteins, offering approximately 100-fold greater sensitivity to tau seeds derived from Alzheimer’s disease brain homogenates than earlier versions. Therefore, this cell line is suited for detecting extremely low levels of tau seeds, critical for early-stage disease research and situations where tau pathology might be subtle and more challenging to detect. Overall, this cell line is useful in screening potential therapeutics that aim to inhibit tau aggregation and supporting studies that track tau pathology over time to study neurodegenerative disease progression.Additional cell lines from this panel: Cat.
