Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. PriGrow IV (TM004) + 50 µM fresh ascorbic acid + 2 nM estradiol + 1 µg/ml insulin + 2.8 µM hydrocortisone + 0.1 mM ethanolamine + 0.1 mM L-glutamine (G275) ,15 nM sodium selenite + 1 ng/ml cholera toxin + 12.5 ng/ml hEGF (Z100139) + 10% fetal bovine serum (TM999) or 35 µg/ml bovine pituitary extract (Hammond) + 1% Penicillin/Streptomycin Solution (G255). GNAO1 Stably Expressing Immortalized Human Mammary Epithelial Cell line expresses GNAO1, the protein of which represent the alpha subunit of the G0 heterotrimeric G-protein signal-transducing complex and is partially responsible for smooth muscle contraction and depressing neuronal activity. They function to inhibit of adenylate cyclase , open K -channels (via β/γ subunits), and close Ca2 -channels. Researchers may use this cell line to study G-protein signal-transducing pathways in humans. Defects in the GNAO1 gene are known as a cause of early-onset epileptic encephalopathy. This cell line was made using retrovirus transduction. It carries flag tag and expresses GFP.
