These novel bone cell lines were established by immortalizing an osteoblastic clonal cell population with avian v-mycOK10 gene. Although both cells are from the same clonal population, T0216 and T0217 have differing properties. T0216 expresses alkaline phosphatase (AP), shows PTH- and PGE2-induced cAMP production, synthesizes mainly collagen type I and a minor fraction of type III, and produces mRNA for the bone-specific protein osteocalcin. Cells are osteogenic, form cartilage and fibrous tissue, and have multipotential differentiation capacity and form bone and cartilage in vivo.
