Featuring a CRISPR/Cas9-induced knockout at the Trp227 site of the PPOX gene, this cell line is devoid of PPOX activity, leading to pronounced accumulation of protoporphyrinogen IX and a halving of heme levels compared to controls. Despite these biochemical alterations, cell proliferation remains unaffected, highlighting the cell’s adaptive mechanisms to heme deficiency.
