PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) are recommended for optimal cell culture. Dulbecco's Modified Eagle Medium (DMEM) ,High Glucose (TM500) + 10% FBS + 1% Penicillin/Streptomycin Solution (G255) ,37.0°C ,5% CO₂. 2.0 mg/ml Puromycin (G264) for selection. Note: Cells may take 48 hours to attach to the vessel. Selection drugs should be added to the culture medium after the first passage to ensure cells have recovered from freeze-thaw conditions. |

AC3 and AC6 encoded by the ADCY3 and ADCY6 genes are the most abundant adenylyl cyclase (AC) isoforms in HEK293 cells. ACΔ3/6 Stable Knockout HEK293 Cell Line was generated by knocking out the AC3 and AC6 genes through stable transfection then selection with puromycin (2 mg/ml) and where then determine to exhibit low cAMP levels.
