The Protrudin-expressing hTERT-RPE1 stable cell line was developed by genetically engineering human retinal pigment epithelial (RPE) cells to overexpress Protrudin. This stable cell line was characterized through various assays, including confocal microscopy and Western blotting, to confirm the expression and functional role of Protrudin. These cells are used in research to study phagocytosis, specifically focusing on the role of Protrudin in mediating ER-endosome contact sites, facilitating the translocation of endosomes to the phagocytic cup, and enhancing the uptake of apoptotic bodies. Unique features include the enhanced formation of phagocytic cups and increased efficiency in the uptake of phosphatidylserine-coated particles.
