This HeLa cell line features a CRISPR/Cas9 mediated stable knockout of C9orf72 which binds to SMCR8, localizes to lysosomes, and plays a critical role in regulating mTORC1 signaling. The knockout of C9orf72 revealed phenotypes such as abnormally swollen lysosomes and altered mTORC1 signaling responses, highlighting its importance in lysosomal function and cellular homeostasis.
